Debug slow nginx wordpress-multisite

Got some issues with my new wordpress-setup being insanely slow.
Storing post are slow, sub-domainname-resolving is slow and sometimes there is a gateway timeout.
This is the setup:
NGINX, Wordpress MU, memcache, mysql, php5.
I've installed a couple of plugins, but these have been deactivated while I was trying to debug.
mySQL is on a single server.
NGINX and the rest is on the main server.
Need some real skills here!
Take apart the whole server-setup and find out what is issues, NGINX conf, php-fpm-conf , the "works"...
Got some skills with Varnish Cache as well?
That would be great! After these issues are resolved I want to setup a varnish on-front, on another instance caching the instance with these issues.
This would be another project.
